摘要

E2F8(E2F transcription factor 8, E2F8)作为含双DNA结合域的非典型E2F转录因子,在肝脏中特异性高表达。生理状态下与E2F7协同调控细胞周期,介导肝细胞多倍体化,维持肝脏稳态。该因子在肝癌、食管癌、胃癌等消化系统肿瘤中高表达,表达水平与患者临床分期、不良预后密切相关,且在肝癌中呈现阶段依赖性双重功能;同时还参与非酒精性脂肪性肝病、肝纤维化的病理调控,有望成为消化系统疾病的诊断标志物或治疗新靶点。

Abstract

E2F8 (E2F transcription factor 8) is an atypical E2F transcription factor with dual DNA-binding domains, which is specifically highly expressed in the liver. Under physiological conditions, it cooperates with E2F7 to regulate the cell cycle and mediate hepatocyte polyploidization, thereby maintaining liver homeostasis. This factor is highly expressed in digestive system tumors such as hepatocellular carcinoma, esophageal cancer and gastric cancer, and its expression level is closely associated with patients' clinical stages and poor prognosis, with a stage-dependent dual function in hepatocellular carcinoma. Meanwhile, it is also involved in the pathological regulation of non-alcoholic fatty liver disease and hepatic fibrosis, and is expected to serve as a diagnostic biomarker or a novel therapeutic target for digestive system diseases.
